Polymorph Energy Landscape: The Seed Lowers the Nucleation Barrier
Why 240 batches held, and what one seed changed
Panel A (Ritonavir): a metastable Form I basin (soluble, therapeutically active) sits beside a deeper Form II basin (more stable, tighter packing, insoluble, inert), separated by a nucleation barrier of about 85 kT; a ball in the Form I basin marks 240 consecutive successful batches, and the red dashed curve shows the same schematic landscape once an epitaxial seed crystal is present, the barrier dropping to about 34 kT and the nucleation rate rising by twenty-two orders of magnitude. Panel B (cocoa butter): three basins of increasing depth, Form IV (dull, crumbly, melts at 27 degrees), Form V (shiny, snappy, melts at 34 degrees; highlighted green as the deepest basin compatible with function) and Form VI (more stable still, whitish bloom), with tempering steering the melt into Form V and uncontrolled cooling landing in Form IV.
